Parmi les microalgues commercialement importantes, la microalgue Haematococcus pluvialis est la source la plus riche et la plus prometteuse d astaxanthine naturelle. Cependant, la culture des microalgues en photobioréacteurs (PBRs) souffre généralement d une perte au niveau de la productivité, en raison des limitations du transfert de la lumière au sein de la culture et du besoin d appliquer un protocole en deux étapes (phase verte de croissance et phase rouge d accumulation d astaxanthine). Afin d améliorer cela, l analyse du transfert radiatif a été effectuée pour étudier son rôle dans le processus. La première partie de cette thèse consiste à déterminer les paramètres spécifiques de la photosynthèse qui, combinés à une nouvelle méthodologie théorique, ont permis d optimiser la production en phase verte. La deuxième partie s est focalisée sur la compréhension du rôle du transfert radiatif dans l accumulation d astaxanthine pour des cultures d Haematococcus pluvialis en limitation et carence azotées. Une forte accumulation ainsi qu une productivité importante en astaxanthine ont été obtenues pour une valeur critique de vitesse moyenne d absorption des photons (MRPA). En démontrant que la production d astaxanthine est également limitée par le MRPA au sein du PBR, il a été possible d optimiser les conditions de culture en batch, mais également en continu, ouvrant ainsi la possibilité d une production optimisée en une seule étape de biomasse enrichie en astaxanthine. | ||
330 | |a Microalgae are known for their high potential to accumulate secondary metabolites, which can be used in several fields. Among the commercially important microalgae, the green microalga Haematococcus pluvialis is the richest and most promising source of natural astaxanthin. However, the cultivation of microalgae in photobioreactors (PBRs) generally suffers from low productivity, due to the light transfer limitations within the culture and the need to apply a two-step protocol (green phase (growth) and red phase of astaxanthin accumulation). In order to improve this, light transfer analysis was performed in this work to investigate its role in the process. The first part of this study consists in determining the specific parameters of photosynthesis, which, combined with a new theoretical methodology, have made it possible to optimize the production in the green phase. The second part were focused on understanding the role of light transfer in astaxanthin accumulation for Haematococcus pluvialis grown in nitrogen limited conditions in both batch and continuous modes. A strong accumulation as well as a significant productivity of astaxanthin were obtained for a critical value of mean photon absorption rate (MRPA). By demonstrating that the production of astaxanthin is also limited by the MRPA, it enables to optimize further culture conditions in batch mode, but also in continuous mode, opening the possibility of an optimized production in a single step of an astaxanthin-enriched biomass. | ||
