Summary : | L astate-211 (t1/2= 7.2h, 1 particule a/désintégration) est considéré comme un radioisotope hautement prometteur pour la radiothérapie interne vectorisée alpha des cancers. La méthode actuellement établie pour son introduction sur un vecteur consiste à former une liaison carbone-astate. Toutefois, un manque de stabilité de cette liaison a été souligné lors d essais précliniques et cliniques, entrainant l irradiation de tissus sains. Quelques alternatives ont d ores et déjà été envisagées, parmi elles, l exploitation d une liaison métal- astate. Notre projet basé sur la théorie HSAB vise à exploiter la formation de liaisons fortes entre un métal de transition mou (rhodium, iridium, or, argent) au degré d oxydation +I et At- en tant que base molle. En raison de leur capacité à former des complexes stables avec les métaux de transition de bas degrés d oxydation, les carbènes N-hétérocycliques ont été sélectionnés comme ligand du métal. Dans ce contexte, l objectif du projet a consisté à évaluer la stabilité de complexes marqués à l astate-211 afin de sélectionner la paire métal/ligand la plus prometteuse en vue d évaluations in vivo. Dans un premier temps la préparation de complexes d encombrements stériques et d effets électroniques variables a été réalisée. Puis leurs marquages à l iode-125 et à l astate-211 ont été accomplis. Pour compenser l absence d isotope stable de l astate, une étude par modélisation moléculaire a été réalisée pour guider les expérimentations. Enfin, l évaluation de la stabilité des composés radiomarqués dans des milieux variés a montré des résultats prometteurs avec un complexe d or(I) encourageant ainsi la poursuite de l étude sur ce métal Astatine-211 (t1/2= 7.2h, 1 particle a/decay) is considered as a highly promising radioisotope for targeted alpha therapy of cancers. The currently established method to bind astatine to a vector is to form a carbon- astatine bond. However, a lack of stability of this bond was highlighted by pre-clinical and clinical trials, leading to healthy tissues irradiation. A few alternatives have already been considered. Among them, the formation of a metal-astatine bond. Our project based on the HSAB theory aims to benefit of the strong bonding between a soft metal (rhodium, iridium, gold, or silver) in the +I oxidation state, and At- acting as the soft base. Due to their well-known capacity to form highly stable complexes with low valent transition metals, N-heterocyclic carbenes were selected as ligands. Our purpose was to evaluate the 211At-complexes stability in order to select the most suitable metal/ligand pair for in vivo evaluation. The synthesis of complexes with various steric hindrance and electronic effects was initially considered. Then, 125I and 211At-labelling were set up. A theoretical study was performed to guide the experimentation in order to compensate the lack of stable At isotope. Lastly, stability evaluation of radiolabelled compounds in different medias showed promising results for a gold(I) complex which encourages us to continue the study of this metal |
