Résumé : | Dans le cadre de la politique environnementale européenne, le plan Ecophyto vise la réduction de moitié de l'utilisation des pesticides d ici 2018. Les solutions alternatives comme le biocontrôle qui utilise des mécanismes anti-phytopathogènes naturels provoqués par des organismes divers pour prévenir et lutter contre les maladies des végétaux cultivés sont privilégiées. Ainsi, l'utilisation de souches fongiques et notamment de Trichoderma d'origine terrestre est déjà développée en biocontrôle dans des préparations commerciales. Le milieu marin étant riche en Trichoderma spp., le but de ce travail a été d'étudier l'intérêt potentiel de ces souches marines pour une utilisation en biocontrôle. Les Trichoderma produisent notamment des petits peptides à synthèse non ribosomale : les peptaïbols. Différents auteurs ont avancé l'hypothèse du rôle important quils joueraient dans l'activité antifongique des Trichoderma qui les produisent. C'est la réponse à cette question et son application pour la sélection des souches utilisables en biocontrôle qui constitue la base de ce travail. Les principaux axes de cette étude ont donc été l'identification de nouveaux peptaïbols, l'évaluation de leur activité antifongique sur des phythopathogènes ainsi que leur quantification dans les extraits de cultures de Trichoderma afin de développer des critères pour une sélection pertinente des meilleurs candidats pour une utilisation en biocontrôle. Aux cours de ces travaux, 2 nouvelles classes de peptaïbols ont été isolées et séquencées, l'action antifongique des peptaïbols longs a été démontrée in vitro et in planta et une méthode de quantification LC-MS a été développée et validée avec succès. Within the European environmental policy framework, the Ecophyto plan targets the half reduction of the pesticides use before 2018. Alternative ways such as the biocontrol that uses natural anti-pathogen mechanisms caused by various organisms are favored. Indeed, the use of fungal strains, particularly those belonging to the genus Trichoderma is already effective in biocontrol with various commercial preparations. Since the marine environment is rich in Trichoderma spp., the aim of this work was to study the potential value of these marine strains or use biocontrol. The Trichoderma spp. produce numerous metabolites including small peptides with a non-ribosomal synthesis : peptaibols. Different authors have hypothesized the important role that these peptaibols play in the antifungal activity of Trichoderma strains with produce them. The answer to this question and its application for the selection of suitable biocontrol strains forms the basis of this work. The main axes of this study were therefore research and the identification of new peptaibols, evaluation of their antifungal activity on phyto-pathogenic fungi affecting crop with a significant economic interest and their quantification in the extracts from Trichoderma cultures to develop criteria for an appropriate selection of the best candidates for biocontrol. In the course of this work, two news peptaibols classes have been isolated and sequenced, the antifungal action of long chain peptaibols has been demonstrated in vitro and in planta a LC-MS method has been developed for their quantification and successfully validated. |
