TY - THES A1 - Berger Jean-Mathieu AB - L'objectif de cette thèse a été d'apporter une base moléculaire aux mécanismes mis en jeu dans l'excrétion transintestinale du cholestérol (TICE). Cette voie, complémentaire à la voie biliaire, permet l'efflux du cholestérol plasmatique directement à travers la muqueuse intestinale. Au cours de ma thèse, J'ai mis en évidence que le TICE est un processus métabolique actif sensible à la température et à l'oxygène. J'ai démontré in vivo et ex vivo chez la souris mais également pour la première fois ex vivo chez l'homme, qu'à la fois les lipoprotéines plasmatiques de type LDL (Low density lipoprotein) ou HDL (High density lipoprotein) peuvent fournir du cholestérol au TICE. Bien que le LDLR (récepteur au LDL) ne soit pas indispensable au TICE, la modulation de son expression affecte le TICE. Ainsi, l'absence de PSCK9 (Proprotein convertase subtilisin / kexin type 9), un inhibiteur endogène du LDLR, ou un régime enrichi en statines augmentent l'expression intestinale du LDLR et stimulent le TICE. Enfin, j'ai également montré que le TICE est inductible par la diosgénine, un stéroïde végétal connu pour ses propriétés hypocholestérolémiantes. Les mécanismes moléculaires impliqués restent néanmoins à être identifiés. Au niveau apical, le transporteur multidrogues ABCB1 participe avec l'hétérodimère ABCG5/G8 à l'efflux entérocytaire du cholestérol. Mes travaux de thèse ont contribué à mieux comprendre les mécanismes moléculaires impliqués dans le fonctionnement et la régulation de cette voie. Sa caractérisation pourrait permettre à terme d'ouvrir de nouvelles perspectives thérapeutiques pour lutter contre l'hypercholestérolémie et les maladies cardiovasculaires chez l'homme. AB - The aim of this thesis was to provide a molecular basis for the mechanisms involved in transintestinal cholesterol excretion (TICE). This pathway, complementary to the biliary pathway, allows the plasma cholesterol efflux directly through the intestinal mucosa. During my thesis, I have shown that TICE is an active metabolic process, temperature and oxygen -sensitive. I have demonstrated in vivo and ex vivo in mice but also for the first time ex vivo in humans, that both types of plasma lipoproteins LDL (Low density lipoprotein) and HDL (High density lipoprotein) can provide cholesterol for TICE. Even if the LDLR (LDL receptor) is not essential for TICE, its expression modulation affects TICE. Thus, the absence of PSCK9 (Proprotein convertase subtilisin / kexin type 9), an endogenous inhibitor of the LDLR or a statins-enriched diet, increase LDLR intestinal expression and stimulate TICE. Finally, I also found that TICE is inducible by diosgenin, a plant steroid known for its cholesterol-lowering properties. However, the molecular mechanisms involved remain to be identified. At the apical level, the multidrug transporter ABCB1 is involved with the heterodimer ABCG5/G8, in the enterocyte cholesterol efflux.During my PhD training, I have contributed to a better understanding of the molecular mechanisms involved in this pathway. Its characterization could eventually lead to open new therapeutic perspectives in the struggle against high cholesterol and cardiovascular disease in humans.
