TY - THES A1 - Baud'huin Marc AB - L'os est un tissu conjonctif très vascularisé siège d'une activité métabolique importante. Les vaisseaux qui colonisent l'environnement osseux apportent les facteurs nécessaires à la différenciation, l'activation et la survie des cellules osseuses. L'homéostasie vasculaire et osseuse repose sur un réseau complexe de messagers solubles (OPG, RANKL, VEGF ) capables d'agir sur les deux systèmes. Nous décrivons, grâce à nos travaux de recherche, l'implication de deux molécules de l'homéostasie vasculaire dans la différenciation ostéoclastique. Le complexe FVIII/Facteur de von Willebrand est capable d'inhiber l'ostéoclastogenèse en s'associant à RANKL et en renforçant l'activité inhibitrice de l OPG. Ce complexe contrôle également la survie cellulaire de cellules d'ostéosarcome et de sarcome d'Ewing en empêchant l'activité protectrice de l'OPG vis-à-vis de la cytokine pro-apototique TRAIL. L'héparine ainsi que l'ensemble des glycosaminoglycanes inhibent la différenciation ostéoclastique en diminuant l'adhérence des précurseurs ostéoclastiques. Ils empêchent également l'étalement des ostéoclastes matures à leur support. Cette étude pourrait permettre de comprendre la mise en place des atteintes osseuses liées à une altération du système vasculaire (hypercoagulabilité liée au processus tumoral et hémophilie) et des lésions vasculaires dans lesquelles les cellules osseuses sont impliquées (athérosclérose et calcification artérielle). AB - Bone is a connective tissue highly vascularized with an important metabolic activity. Blood vessels which colonize the bone environment bring the factors necessary to the differentiation, the activation and the survival of the bone cells. A complex network of soluble messengers (OPG, RANKL, VEGF ) controls vascular and bone homeostasis. We describe, through these studies, the implication of two molecules of vascular homeostasis. FVIII/von Willebrand factor complex is able to inhibit osteoclastogenesis by binding to RANKL and by reinforcing the inhibitory effect of OPG. This complex controls also cell survival of osteosarcoma and Ewing s sarcoma cells by preventing the binding of OPG to the pro-apoptotic cytokine TRAIL. Heparin and the whole of glycosaminoglycans inhibit osteoclast differentiation by decreasing the adhesion of osteoclast precursors. They also inhibit spreading of mature osteoclasts. Our results complete the molecular network shared by bone and vascular systems. This study could make it possible to understand the establishment of the bone lesions induced by a deterioration of the vascular system (hypercoagulability due to tumoral process, hemophilia) and vascular injuries where bone cells are implied (atherosclerosis and arterial calcification).
